A beginner’s guide to surface plasmon resonance

چکیده
Surface plasmon resonance (SPR) has emerged as a powerful optical detection technique for studying the binding behaviour of immobilized ligands and analytes in solution. The makes it possible to measure interactions real time with high sensitivity. Over past two decades, SPR become gold standard biomolecular biomedical research drug discovery. that can be studied are diverse include protein–protein, protein–small molecule, protein–nucleic acid, protein–carbohydrate, lipid–protein, hybrid systems biomolecules non-biological surfaces. allows researchers determine which molecules interact, how strongly they bind inform experiments using mutants, truncations or other variations probe specificity. This article summarizes principle experimental set-up illustrates utility example lipid–protein interactions.

Introduction to Surface Plasmon Resonance
Since its first observation by Wood in 1902 [1,2], the physical phenomenon of surface plasmon resonance (SPR) has found its way into practical applications in sensitive detectors, capable of detecting sub-monomolecular coverage. What is surface plasmon resonance? Wood observed a pattern of ‘‘anomalous’’ dark and light bands in the reflected light, when he shone polarized light on a mirror with ...

متن کاملImaging surface plasmon resonance
The central theme of this thesis is the use of imaging Surface Plasmon Resonance (iSPR) as a tool in the characterization of surfaces with laterally varying properties. Within the scope of this work, an instrument for iSPR analysis was designed and built. SPR is a very sensitive technique for monitoring changes in optical properties in the immediate vicinity of a sensor surface, which is very u...
